Resources - STEDCore (CBIA's SuperResolution Suite)

 

STEDCore is the first super-resolution Stimulated Emission Depletion Continuous Wave (STED CW) research platform in Canada. The platform was developed by Dr. Steffany Bennett with the generous support of two Canadian Foundation Innovation (CFI) grants (CFI#16065 and CFI#23890). STEDcore is part of the Ottawa Hospital Research Institute (OHRI) Imaging and the uOttawa/Faculty of Medicine Cell Biology and Image Analysis (CBIA) Facilities. The Bennett laboratory runs STEDCore within CBIA as a collaborative research platform available to all researchers. The platform, both confocal and super-resolution systems, is operated by Mr. Mark Akins who works with Dr. Bennett to provide researchers with imaging expertise, obtain all images, calibrate and maintain all of STEDCore's equipment, develop new super-resolution analysis protocols and, if requested, perform critical experiments for users. All imaging is down by Mark Akins with users as per our agreement with Leica and the terms of our service contract.

Lasers:

STED CW depletion laser: 592 nm
Note: Only fluorophores with excitation below the 592 nm wavelength and emission passing through 592 nm can be used (aka green STED only).  The list of fluorophores we have optimized extensively are listed on our secure site for STEDcore members.

 

There is a minimum 1 hr charge. Imaging occurs in afternoon.

 

There is a $50.00 set-up fee to align and verify the STED CW depletion laser in the AM before experimental analysis (to ensure STED CW is optimal prior to researcher analysis). There are days we are not able to obtain super-resolution due to temperature, humidity, construction (vibrations) etc. Investigators will be notified by noon as to whether the system can be used that afternoon. If conditions are sub-optimal, the session will be canceled. Users will not be charged. Users will be rebooked once the system has been verified. If conditions are optimal, users are charged the $50.00 setup fee and notified that the afternoon session is confirmed. STED imaging can be scheduled Tuesday, Wednesday, or Thursday. To book a time please email Mark Akins at stedcore@uottawa.ca. 48 hours notice must be given.
